Our Port access only in VPN and internal Network

We need to access our server via VPN and internal Network only, no need to access an external network,

Please suggest the following steps to access our server port.

I suggest setting up corresponding firewall policies to achieve your desired goal. It is definitly possible to do that with a FortiGate.
